Hi,
My Instance Definitions’s layer is the Layer 01, I add two blocks by Layer 01, and then change one of them to Layer 02, after I hide the Layer 01, the block in Layer 02 still hide too.

Hello- both the definition geometry layer(s) and the layer that the instance is on must be visible to see all objects in a block.
In your file, the smaller block has the definition on Layer 01 and the instance is on Layer 02; the larger one has both instance and definition on Layer 01.
