Can you turn off layers in multiple layouts or is the only option to turn off one layout at a time using the Layouts side menu?

If you are asking about layers that have objects ON the layout, then, no, you’ll need to turn off that layer in each layout.
If, on the other hand, the question is about layers that have objects in model space that you can see in details on the layout, there are a few new options for dealing with those in the latest Rhino 8 WIPs:
